I think the engineers were playing with fire (Black Goo) and got burnt. Could it be Alien (Xeno) DNA? Were the original Xeno's something more latger, frightening, dangerous...

Indeed I believe the true representations of the Promethean myth are the Space Jockeys not only the humans aboard the ship Prometheus. The ship Prometheus is but a folly of Weyland in his pride and arrogance likening himself to the titans taking on the Gods, yet the Space Jockeys are the real Prometheus claiming fire and getting burned... and how.
